Fluzone® Trivalent is an egg-based, inactivated split-virus influenza vaccine indicated for intramuscular injection. It is supplied as a sterile, preservative-free, latex-free injectable suspension in single-dose prefilled syringes.

Fluzone is clear and slightly opalescent in appearance.

Manufacturing Process

Fluzone is produced using traditional egg-based influenza vaccine technology:

• Influenza viruses propagated in embryonated chicken eggs
• Virus-containing allantoic fluid harvested
• Inactivated with formaldehyde
• Concentrated and purified using linear sucrose density gradient centrifugation
• Chemically disrupted with octylphenol ethoxylate (Triton® X-100) to create a “split-virus” formulation
• Further purified and suspended in sodium phosphate-buffered isotonic sodium chloride solution
• Each strain manufactured separately and combined into the final trivalent formulation

Antigen Content

Standardized according to United States Public Health Service (USPHS) requirements.

Each 0.5 mL dose contains:

• 45 micrograms (mcg) total hemagglutinin (HA)
  • 15 mcg HA from each of the three recommended influenza strains

Preservative & Latex Information

• Single-dose prefilled syringes:
  • Preservative-free
  • No thimerosal
  • No natural rubber latex
• Multi-dose vial (5 mL) presentation:
  • Contains thimerosal (mercury derivative)
  • 25 mcg mercury per 0.5 mL dose
  • 12.5 mcg mercury per 0.25 mL dose
